A differential chemoreactome analysis was employed to evaluate the effects of the muscle relaxant tolperisone and five NSAIDs (dexketoprofen, etoricoxib, meloxicam, naproxen, and diclofenac). The biological activities of the molecules under study were assessed in five sections: 1) inhibition of the proteins of prostaglandin and leukotriene metabolism; 2) inhibition of the effects of the transcription factor nuclear factor kappa, tumor necrosis factor-, and other anti-inflammatory mechanisms; 3) inhibition of excessive blood coagulation and platelet aggregation; 4) vasodynamic effects; 5) antitumor properties on cell lines in culture.</p></sec><sec><title>Results and discussion</title><p>Results and discussion. Based on the differences in the pharmacological activity profiles of tolperisone and NSAIDs under study, the investigators identified the most promising synergistic combinations, in which both active ingredients complemented each other as effectively and safely as possible. The obtained estimates of the degree of synergism of various combinations of tolperisone and NSAIDs hold that the most promising antithrombotic, and antitumor effects.</p></sec><sec><title>Conclusion</title><p>Conclusion. The results of this study will help adequately choose combinations of muscle relaxants and NSAIDs in patients with muscle hypertonia, which will be able to improve the efficiency and safety of treatment.</p></sec></abstract><trans-abstract xml:lang="ru"><p>Перспективным направлением терапии болезненного гипертонуса мышц и судорожных состояний является совместное использование миорелаксантов и нестероидных противовоспалительных препаратов (НПВП).</p><p>Цель исследования – определить наиболее эффективные и безопасные синергидные комбинации толперизона и НПВП.</p><sec><title>Материал и методы</title><p>Материал и методы. Проведен дифференциальный хемореактомный анализ эффектов миорелаксанта толперизона и пяти НПВП (декскетопрофен, эторикоксиб, мелоксикам, напроксен, диклофенак). Оценка биологических активностей исследуемых молекул проводилась по пяти разделам: 1) ингибирование белков метаболизма простагландинов и лейкотриенов; 2) ингибирование эффектов транскрипционного фактора NF-kB, фактора некроза опухолей и другие противовоспалительные механизмы; 3) ингибирование избыточной коагуляции и агрегации тромбоцитов; 4) вазодинамические эффекты; 5) противоопухолевые свойства на линиях клеток в культуре.</p></sec><sec><title>Результаты и обсуждение</title><p>Результаты и обсуждение. На основании различий в профилях фармакологической активности толперизона и исследуемых НПВП были выделены наиболее перспективные синергидные комбинации, в которых оба действующих начала дополняют друг друга максимально эффективно и безопасно. Полученные оценки степени синергизма различных комбинаций толперизона и НПВП дают основание утверждать, что наиболее перспективными являются комбинации толперизон + эторикоксиб и толперизон + декскетопрофен, так как именно в этих комбинациях усиливаются противовоспалительные, антитромботические и противоопухолевые эффекты.</p></sec><sec><title>Заключение</title><p>Заключение.
